Rice production in Malaysia is facing challenges such as declining yields due to soil infertility and the environmentally harmful practice of open burning of rice straw. Rice straw is a major agricultural by-product of rice production. Sustainable methods for managing rice straw by utilizing fungi to degrade lignocellulosic biomass and can be produced into biofertilizers. The objectives of this study were to determine the xylanase enzyme activity isolated from rice straw and compare the enzyme activities of different fungus strains through statistical analysis. Seven fungal strains isolated from rice straw were revived by sub-culturing on PDA plates to promote active growth. They were then cultured in Czapek-Dox broth for seven days to production enzymes. The crude enzyme was obtained by centrifuging the culture broth and subsequently used for the xylanase activity assay. Fungus strain 4D demonstrated the highest xylanase activity at 1.1321 U/ml, while fungus strain P3 exhibited the lowest activity at 0.0358 U/ml. Lastly, the best fungal strain of 4D for xylanase was identified in the post-hoc test which has the highest value of mean with the subset of strains that have statistically significant differences in enzyme activity (p < 0.05). In conclusion, the findings underscore the potential of fungal strains, particularly strain 4D, in rice straw degradation, which can contribute to sustainable agriculture and environmental conservation.
