| Summary, etc. |
Cyclodextrin glucanotransferase (CGTase) has an ability of degrading starch to produce cyclodextrin (CD). CD has a high demand in various industries such as food industries, pharmaceutical industries, textiles, and environmental engineering. The instability and denaturation of CGTase lowers the production of CD during the reaction process. Therefore, enzyme immobilization method is the best approach to overcome the problems. Production of CD by the immobilized CGTase is based on a few process parameters such as contact time, pH and temperature. In this present study, the effect of pHs (5, 6, 7, 8, and 9), temperatures (20, 25, 30, 35, 40 °C) and agitation rates (50, 100, 150, 200, 250 rpm) on the CD production using immobilized CGTase from Bacillus licheniformis on banana peel was studied. The highest production of CD was observed at pH 7 (3.74 mg/mL of CD), temperature 25°C (3.10 mg/mL of CD) and 150 rpm of agitation rate (3.13 mg/mL of CD). Banana peel waste selected as support consisting cellulose abundantly, thus they are capable for binding enzymes for immobilization process. Moreover, banana peel is an ecologically harmless and low-cost material. Thus, it could be concluded that the immobilized enzyme could produce high CD production with high enzyme stability banana peel is an effective support for the high production of CD on an industrial scale.
