The present study will discuss the processing attribute effects on granule size distribution due to high-shear wet granulation, which is an important unit operation within pharmaceutical tablet manufacturing. High-shear wet granulation is generally used to enhance flowability, compressibility, and uniformity of powders in tabletting. These include the four major factors: binder type, binder concentration, impeller speed, and chopper speed, which are systematically studied in this paper using the DOE approach. Riboflavin was used as an active pharmaceutical ingredient, and polyvinylpyrrolidone (PVP) and hydroxypropyl methylcellulose (HPMC) were binders. The granules were evaluated for size distribution by sieving, while post-compression analysis included weight variation, hardness, friability, disintegration, and dissolution tests following USP Pharmacopeia specifications. Results showed that binder type and concentration had the most profound influence on granule size and uniformity. Larger and more uniform granules were produced with PVP compared to HPMC. Impeller and chopper speed also played a critical role, particularly in balancing granule size and reducing fines. The optimal conditions improved tablet characteristics like faster disintegration and higher mechanical strength. This study underlines the importance of optimization of processing parameters to enhance product quality, reduce material waste, and support sustainable manufacturing. The results provide valuable insights into the pharmaceutical industry, especially in the development of high-shear wet granulation techniques for better efficiency and consistency in the production of tablets. |
