Congenital Adrenal Hyperplasia (CAH) refers to genetic disorder of the adrenal glands. People with CAH are unable to make enough cortisol and aldosteron hormone in their adrenal glands, hence imbalance of hormones occur. This research is about to increase the production of immunoglobulin G (IgG) in the production of monoclonal antibody (MAb) for CAH diagnostic kit. The purpose of this research is to screen and optimize the potential inducers in maximizing the production of IgG by hybridoma 192 cell line. There are six (6) inducers used in this research, they are Lysozyme, Aldolase, Sodium Butyrate, Sodium Phosphate, Potassium Phosphate and Non-essential Amino Acid. Hybridoma 192 cell was cultured in 5% CO2 incubator, at temperature of 37°C and greater than 80% humidity in the medium with different concentration of inducers agent. The samples were taken after 72 hours of culture. Productions of Mabs were screened using Enzyme-linked Immunosorbent Assay (ELISA) to determine the inducers capability in increasing IgG production. Method is continued with optimization using Response Surface Methodology (RSM). Two parameters are selected; optimum concentration of inducer agent and time of addition the inducer agent. From the result from screening of inducers, sodium phosphate was found as the most capable inducing agent to stimulate IgG production at a concentration of 25mM. Thus, optimization was only focus on manipulating the concentration in the range of 20mM to 30mM and the addition time of sodium phosphate. At the end of optimization by RSM, the result shows that time of sodium phosphate addition do not have any significant effect to the production while the increasing of the concentration of sodium phosphate will only decrease cell activity and reduce the production of IgG. In a nut shell, the optimization stage was unsuccessful and for further research, low concentration range should be considered. |
