Summary, etc. The composition of oil palm trunk sap (OPTS) as a fermentation feedstock to produce bioethanol by using Saccharomyces cerevisiae was investigated. The effects of nutrient addition namely, ammonium sulfate ((NH4)2SO4), disodium hydrogen phosphate (Na2HPO4), magnesium sulfate (MgSO4) and L-alanine (C3H7NO2) were known to improve yield product and hence were investigated thoughout this study. The OPTS was obtained from a 30 years old oil palm trunk. Bioethanol fermentation from OPTS was conducted by using Baker’s yeast, with and without nutrient supplementation. The sugar content in OPTS and fermentation mother liquor was determined using a high-performance liquid chromatography (HPLC). Analysis of OPTS composition indicated glucose as a dominant sugar with concentration of 75.51 g/l, followed by sucrose (62.68 g/l) and fructose (29.41 g/l). The OPTS contained 172 g/l total fermentable sugar. The maximum value of theoretical bioethanol yield was 51.08% from OPTS fermentation without nutrient addition. Nutrient addition improved the yield markedly with the highest bioethanol yield as 85.32%, achieved by adding (MgSO4) + (C3H7NO2). The nutrient addition increased the ethanol concentration from 28.76 g/l to 35.64 g/l. The rank of single nutrient influence on improving bioethanol yield was MgSO4 > C3H7NO2 > (NH4)2SO4 > Na2HPO4. However, the combination of (MgSO4) + (C3H7NO2) was higher than any single nutrient addition. Optimisation of parameters including initial pH, temperature and agitation rate was performed by experimental design using response surface methodology (RSM) with applied rotatable central composite design (CCD). Validation of experiment model showed that the optimum conditions for response of actual bioethanol yield (75.82%) can be achieved at initial pH (5.79), temperature (31.05C), and agitation rate (164.38 rpm). The results was within 2.44% of percentage error margin from the predicted result. The supplement employed in this work showed an enhancement of ethanol production from OPTS without complicated pretreatment. The effects of nutrients supplementation and optimization of parameters for bioethanol production using fermentation process from OPTS by Saccharomyces cerevisiae were successfully carried out in this study.
