This study aimed to extract and characterize the antimicrobial properties of Fenugreek, and to fabricate and characterize Fen/PLA/Gel/NCC nanofibers. Fenugreek extract was obtained through maceration and characterized using UV-Vis spectroscopy, confirming the presence of antimicrobial compounds such as phenolic acids, flavonoids, and trigonelline. Nanofibers were fabricated using needleless electrospinning, with the process parameter of height between wire electrode and aluminium foil optimized to produce bead-free nanofibers. Advanced characterization techniques, including SEM, FTIR, TGA, and XRD, were utilized to analyze the morphology, thermal stability, and crystalline structure of the nanofibers. Results demonstrated that the optimized Fen/PLA/Gel/NCC nanofibers exhibited enhanced antimicrobial efficacy against Gram-positive and Gram-negative bacteria, as evidenced by significant inhibition zones. The incorporation of NCC improved the thermal stability of the nanofibers, while the combination of PLA, Gelatin, and Fenugreek extract contributed to their biocompatibility and biodegradability. The best-performing nanofiber was identified with the ratio of Fen/PLA/Gel/NCC at 10:10:0.2:0.1 % w/v, providing the optimal balance of antimicrobial activity, structural integrity, and thermal stability. Recommendations include further optimization of fabrication parameters, comprehensive antimicrobial testing, drug efficiency studies, and isolation of specific bioactive compounds from Fenugreek extract to maximize the nanofiber’s potential. This study underscores the feasibility of Fen/PLA/Gel/NCC nanofibers as an effective solution for antimicrobial applications in healthcare and material science.
