This research investigates the application of enzymatic hydrolysis of mannanase in poultry feed to improve nutritional efficiency and promote the health of poultry. Mannan, a complex polysaccharide found in various feed ingredients, poses challenges to poultry digestion due to its anti-nutritional properties. This study aims to investigate experimentally the effect of reaction parameters on enzymatic hydrolysis of poultry feed using mannanase. To investigate the effectiveness of mannanase supplementation, a series of experiments were conducted using different concentrations of mannanase (6% - 20%) in poultry feed, enzyme (Mannanase) loading (4mL - 20mL) used and the reaction time (30 - 150 minutes). The variables obtained by using One Factor analysis from Design Expert 7 software. An analytical technique, enzyme activity assays were employed to assess the mannose content in terms of its percentage in the poultry feed where the poultry feed is reacted with phosphate buffer and the mannanase enzyme. The result for every variable is produced by maintaining the other variable to achieve the current variable. Based on the findings, the variable that produces the highest amount of mannose was selected as the ideal parameter where 6% of substrate concentration (0.2535 mg/mL of Mannose), 18mL (0.5614 mg/mL of Mannose) and 90 minutes (0.5583 mg/mL of Mannose) is selected. We can conclude that the reaction of the feed and the enzyme acts as a lock and key that dependent among themselves. The reaction will become stable when the total of enzyme or feed is fully reacted. The implementation of mannanase supplementation in commercial poultry production holds significant potential for reducing production costs, increasing sustainability, and meeting the growing demand for high-quality poultry products. For future investigations, it is proposed that the range be narrowed to the high increment of mannose production for each variable to optimize the mannose production.
