Acetic acid has been produced through fermentation through the oxidation of ethanol using Acetobacter spp., known as acetic acid bacteria (AAB). The current challenge in this research lies in identifying the specific requirements for inoculum development of wild strain AAB because to increase the acetic acid production, growth factors need to be optimized using statistical analysis. So, this study will be focuses on varying glucose concentrations to understand the correlation between carbon availability and AAB growth, followed by the characterization of inoculum. To achieve this objective, a wild strain of AAB isolated from MD2 pineapple waste will be used to explore the growth characteristics and suitability for inoculum development. The investigation will focus on the impact of varying glucose concentrations (30, 50, 60) in g/L as the carbon source on the growth of AAB cells and further undergo a detailed characterization, including key parameters such as motility, Gram staining, and the potency index (PI). The results indicate that the optimal sugar concentration for the experiment is at 50 g/L, where the OD is 0.4065, glucose consumption is 27.5, and the PI value is 1.83. The inoculum, grown in the media, produced a Gram-negative coccobacillus (rod shape) with no specific arrangement under these favourable conditions. These characterization methods arc hosed for their relevance evaluating the quality of the inoculum.
