Poultry feed such as corn contained the complex structure of mannan . Mannanase hydrolyzed poultry feed to simpler molecule thus reducing the sugar mannose. Mannanase have been used to improve feed efficiency and utilization in poultry.Improved feed can decrease the level of certain nutrients in the diet while simultaneously lowering the amount of nutrient excretion into the environment, thereby diminishing the risk of eutrophication and acidification from fecal matter. This research aimed to investigate the effect of reaction parameters pH (5,6,7,8,9) ,time (30, 65,100,135,161, 188,214,240) minutes, temperature (40,47, 53,60,65,70,75, 80)°C on enzymatic hydrolysis of poultry feed using mannanase. Reacting poultry feed with mannanase, heated in water bath followed by centrifugation and then dinitrosalicylic acid (DNS) solution is added to terminate the reaction. After that the absorbance of the solution is measured using a UV-Vis spectrophotometer at 540nm. The best results from this research is pH 6 (0.0306mg/mL) , 65 minutes of time (0.0218mg/mL) and 60ºC (0.0345mg/mL) on enzymatic hydrolysis of poultry feed using mannanase Based on the findings, this research concludes that enzymatic hydrolysis of mannanase in poultry feed represents enhance the health of poultry and help poultry feed industry to get the best condition in making poultry feed for improve growth performance of poultry.
