Quercetin is a bioactive flavonoid present in different types of plant products, which is famous for antioxidative, anti-inflammatory, and therapeutic actions. However, due to its insolubility in water and very poor bioavailability, its effectiveness as an oral health supplement remains very low. The purpose of this study is to formulate and characterize a quercetin tablet formulation employing polysorbate 20 as a solubility-enhancing excipient. The objectives include determining the solubility, dissolution rate, and physical properties of quercetin tablets following the impact of using varying concentrations of polysorbate 20 at concentrations of 2%, 2.75%, 3.5%, 4.25%, and 5%. The technique includes wet granulation to obtain granules and tablet compression through a single-punch tablet press. Tablet friability, hardness, disintegration, and dissolution tests are performed to check the quality and efficacy of the tablets which are in conformities to USP standards. The focus of this research is to overcome the solubility issues associated with quercetin by formulating a higher-yielding method that improves its pharmacological value. Finally, the best composition of polysorbate 20 for formulation of quercetin tablets was 14% of polysorbate 20. In conclusion, this study provides knowledge in the enhancement of oral drug delivery systems of poorly soluble compounds for better therapeutic outcomes and patient compliance.
