Nigella Sativa contained abundance of active ingredient useful for anticancer, antibacterial, anti-inflammatory, among others. Effectiveness of nutraceutical products in preventing diseases depend on the bioavailability of the active ingredients. Study was done to investigate the composition of bioactive compounds from Nigella Sativa essential oil using different methods. Extraction of essential oil from Nigella Sativa seeds using maceration solvent extraction and ultrasonic-assisted extraction (UAE) was explored. The effect of different solvent (n-hexane, ethanol, methanol, 50% ethanol, 50% methanol) on the composition of essential oil (EO) yield was investigated using maceration solvent extraction. EO obtained was further analysed using GC-MS. It was indicated that ethanol is the best solvent by taking consideration component extracts and safer to be used. Palmitic acid (7.33-10.17%), linoleic acid (4.95-76.82%) and oleic acid (0.07-13.19%) are the major components EO extracted using pure solvents. The effect of different time, frequency, power and temperature were carried out using UAE method. The extraction conducted at 30 min, 50ᴼC, 53 kHz and 88% power showed the highest value for TPC and TFC which are 57.97 mg GAE/100g sample and 178.305 µg Q/100g sample respectively whereas 63.13% inhibition of DPPH was shown. The results indicate that Nigella Sativa EO can be obtained in a simple technology, faster and low cost by using UAE method.
