  <abstract>Poly(3-hydroxybutyrate), P(3HB) or Polyhydroxybutyrate (PHB) was a common biopolymer plastic of PHA which accumulate as intracellular product of bacteria The study had been conducted on the cheap raw materials including by-product of industrial process and waste of agriculture including of Oil Palm Trunk Sap (OPTS). The purpose of this experiment was to study  the production of PHB biopolymer by using Oil Palm Trunk Sap (OPTS) as carbon source which was glucose. The processes involved with fermentation process, the analyzed on the cell dry weight, analyzed on PHB and optimization process. The fermentation was done in the shake flask with the combination of percent of Oil Palm Trunk Sap (OPTS), inoculum 2, ultrapure water and Mineral Salt Medium (MSM) with working volume of 200 mL. Then, manipulated on the temperature, the agitation speed and the percent of Oil Palm Trunk Sap by using a table of the factorial design. Next, the cell dry weight or biomass and PHB were analyzed for continuity of optimization process. In optimization process, the area contained maximum yield was calculate depend on the three parameters which were temperature, agitation speed and percent volume of Oil Palm Trunk Sap by using Yates’ method and Linear Regression after factorial design. After experiment was finished, the highest production for cell dry weight (g/L) was 5.55 g/L meanwhile the production for PHB was 2.935 g/L. The parameters for both result were 29.6⁰C of temperature, 168 RPM of agitation speed and 28.8 per cent of volume of Oil Palm Trunk Sap. In optimization, for Yates’ method, the main effect was agitation speed and interactive effect was combination between temperature and percent volume of Oil Palm Trunk Sap for both cell dry weight and PHB concentration. Lastly, by using Linear Regression, the data was confirmed contain maximum yield at area investigated.</abstract>
