520 3 _aThe incidence of Type 1 diabetes mellitus (T1DM) has increased globally over the years. Accumulating research suggests that the human complement system may contribute to the pathogenesis of diabetes and induce vascular complications that characterize this condition. Several discoveries from countries such as the UK, China and United States indicated a significant level of complement C3 and C3a in T1DM patients compared to healthy individuals. However, no relevant study has been conducted among T1DM patientsin Malaysia. This study investigates the role of complement C3 and C3a in T1DM patients in Malaysia, with the association of phenotypic characteristics such as age group, ethnicity and gender. A total of 18 (9 T1DM patients and 9 health individuals) were involved in this study and blood serum was obtained for both complement C3 and C3a analysis. The human complement C3 sandwich ELISA kit and human complement C3a sandwich ELISA kit were employed to determine both complement C3 and C3a concentrations in blood serum. The Wilcoxon matched-pairs signed rank test with a Pvalue, 0.1641 for complement C3 and P-value > 0.9999 for complement C3a do not demonstrate any appreciable differences among T1DM patient and healthy control. Based on the findings, the complement C3 and C3a level do not associated with phenotypic characteristics (age, gender and ethnicity) among T1DM patients and healthy controls. As evidence, both Wilcoxon matched-pairs signed rank test (gender) and Kruskal-Wallis tests (age group and ethnicity) were run among T1DM patients and healthy control. P-values obtained (0.3828, 0.1070 and 0.1341) for complement C3 analysis among this group do not show significant differences with these phenotypic characteristics. The results go same with the complement C3a analysis with a P-value of 0.7422 for gender, 0.5283 for age group, and 0.6769 for ethnicity, demonstrating no statistically significant variations based on these phenotypic factors. Therefore, complement C3 and C3a may not be a good indicator in distinguishing between T1DM patient and healthy control in Malaysia.
