520 3 _aBananas (Musa spp.) are a key agricultural crop, offering essential nutrients and significant economic value globally. In Malaysia, different types of bananas are widely cultivated varieties with distinct growth and aromatic characteristics. This study analyzed differential gene expression related to growth and aroma in three Malaysian banana varieties, which are Pisang Mas (Musa acuminata), Pisang Tanduk (Musa paradisiaca), and Pisang Rastali (Musa sapientum). Key target genes investigated were Xyloglucan Endotransglucosylase/Hydrolase (XTH), associated with fruit softening and growth, and Alcohol Dehydrogenase (ADH), critical in aroma compound biosynthesis. Total RNA was extracted from the mesocarp of ripe and unripe samples using the RNA extraction kit, followed by complementary DNA (cDNA) synthesis. Primer sequences for XTH and ADH were designed using data from the GenBank database. Gel electrophoresis confirmed the integrity of the extracted RNA, and UV spectrophotometry verified its concentration and purity. PCR analysis revealed significant variations in gene expression among the banana varieties and across ripening stages. PCR amplification showed that Pisang Mas exhibited robust expression of both XTH and ADH genes across ripening stages. In Pisang Tanduk, XTH expression was prominent, while ADH expression was lower. For Pisang Rastali, both XTH and ADH expressions were comparatively lower than other bananas. This study highlights the potential of leveraging gene expression data for crop improvement, such as enhancing fruit quality, extending shelf life, and improving aroma profiles.
