520 3 _aThis study investigates the impact of key processing attributes on granule flow properties in high shear wet granulation (HSWG) for tablet manufacturing. The four main attributes examined were impeller speed, chopper speed, binder type, and binder concentration. A two-level factorial design was employed to screen and identify the most significant factors influencing granule flowability. The study utilized riboflavin as the active pharmaceutical ingredient (API), with lactose as the filler, HPMC and PVP as binders, and talc and magnesium stearate as glidant and lubricant, respectively. Experimental results demonstrated that chopper speed, binder type, and binder concentration had a significant impact on granule flowability, whereas impeller speed showed a lesser effect. The findings indicated that PVP at a higher concentration (10%) with a low impeller speed and high chopper speed resulted in superior flow properties. Conversely, HPMC required higher mechanical energy for optimal granule formation due to its strong binding properties. The study also highlighted the importance of optimizing granulation parameters to achieve excellent flowability, ensuring uniform die filling and tablet weight consistency. Overall, this research provides valuable insights into the formulation and process optimization of riboflavin granules, contributing to improved tablet quality and manufacturing efficiency. Future studies could focus on refining these parameters and exploring post-compression characteristics, including tablet hardness, friability, and dissolution profiles, to enhance pharmaceutical product performance.
