520 3 _aThis study was carried out to find a new possible halal source of glycosaminoglycans (GAGs) for potential anticancer agent. The under-valued and rarely-bought cartilaginous Blue-spotted stingray was selected for the extraction and characterisation of GAGs. The objectives of the study are to isolate and extract the GAGs from the selected stingray and to characterise the GAGs by using characterization assay, FT-IR and NMR. Extraction was conducted to get the crude of sulfated GAGs from the Blue-spotted stingray. The crude GAGs obtained from the extraction was characterized by using Blyscan and Carbazole quantification assays. Blyscan assay result showed that the total sulfated GAGs amount obtained from the total crude GAGs is 3.9628 g. Quantification by carbazole assay strongly suggested that the crude extract contains 4.7480 g of uronic acids. The crude extract was then analysed by using Fourier Transform Infrared Resonance (FT-IR) and Nuclear Magnetic Resonance (NMR) to confirm its functional groups and molecular structures. From the FT-IR and NMR analysis, the spectra showed the presence of glycosaminoglycans in the crude extract of Blue-spotted stingray. The anticancer property of the crude extract was then studied by using human adenocarcinoma A549 (lung cancer). The results showed that crude GAGs can inhibit the A549 cells depends on the concentration used. As conclusion, blue-spotted stingray can be a halal source of potential GAGs as the yield of the crude GAGs extracted is comparable to the previous research findings. Furthermore, it showed cytotoxic property as it can inhibit the cancer cells growth. In future, the extracted GAGs from blue-spotted stingray could be a promising source of anticancer agent.
