Polyhydroxyalkanoates (PHA) are linear polyester produced in nature by bacterial fermentation of sugar or lipids that was first discovered by Lemoigne in 1925. Poly-Β-Hydroxybutyrate (PHB) is the most well known member of the PHAs. PHB is natural, biodegradable polyester, which is accumulated in the form of intracellular granules by a large variety of bacteria. These granules act as energy reserve materials when nutrients such as nitrogen and phosphorus source are available in limiting concentration in the presence of excess carbon source. This research studies the effect of variables in optimizing the production of PHB from bacteria Cupria vidus necator. In this study, the variables that being studied is temperature (28°C-35°C), glucose concentration (10g/L – 30g/L) and agitation speed (150rpm-300rpm). The purpose of this study is to find the value of media variable that will optimize the production of PHB in the fermentation process of biopolymer production. From the experiment and calculation, the maximum yield of biomass and PHB is achieve at the temperature of 34C, glucose concentration of 15.63g/L and at agitation of 222.83rpm. The maximum predicted yield for biomass (cell dry mass) is 3.07855g/L and the maximum predicted yield for PHB is 0.564093g/Lo. -Author
