This study investigates the fabrication and drug release kinetics of polylactic acid (PLA)-starch-nanocrystalline cellulose (NCC) nanofibers loaded with paracetamol, developed using the electrospinning technique for biomedical applications. The primary objective is to study the controlled release of paracetamol and evaluate the suitability of different kinetic models in describing drug release behavior. Various concentrations of starch, NCC, and paracetamol were incorporated into the PLA matrix to determine their effects on nanofiber properties. The nanofibers were characterized using Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM), and UV-Visible Spectroscopy (UV-Vis). Swelling and degradation tests assessed the stability and drug release performance of the nanofibers over time. The drug release kinetics were analyzed using zero-order, first-order, and Higuchi models to determine the most accurate fit. The results indicated that the addition of NCC improved the structural integrity of the nanofibers, while higher paracetamol concentrations accelerated degradation and swelling. The Higuchi model best described the drug release behavior at lower paracetamol concentrations, whereas a combination of zero-order and first-order models provided a better fit for higher concentrations. This study demonstrates the potential of PLA-starch-NCC nanofibers as a sustainable and effective transdermal drug delivery system. Further research should explore in vivo testing and alternative biopolymer formulations to enhance drug release efficiency and biocompatibility. |
