The perennial herb, family Zingiberaceae, Curcuma longa, is famous for its bioactivities and its use in its curative activity and its use in modern wellness supplements. Traditionally, it is for its curative activity and an integral part in modern wellness supplements. The bioactive compound in the herb, curcumin, has a variety of actions including antibacterial, anti-inflammatory, and antioxidant activity. Despite its medicinal value, most claims have not yet been substantiated by solid scientific research, especially when it comes to maximising its extraction for the purpose of maximising its bioactivities. In this study, the effectiveness and bioactivities of Ultrasonic-Assisted Extraction (UAE) for Curcuma longa extract extraction are studied. Antioxidant, anti-inflammatory, and antibacterial activity of extract were evaluated in an endeavour to evaluate its suitability for use in drugs. Antioxidant activity of the extract and ascorbic acid was evaluated through H₂O₂ free radical scavenging activity with an IC50 value of 295 μg/mL and 75 μg/mL respectively. This shows that theantioxidant activity of extract is considered poor. Anti-inflammatory activity of extract and sodium diclofenac was evaluated through BSA denaturation assay which exhibited significant activity with an IC50 value of 213.79 μg/mL and 175 μg/mL respectively. Antibacterial activity was evaluated through disc diffusion testing with Staphylococcus aureus and Escherichia coli. Antibacterial activity with significant activity towards Staphylococcus aureus (10 mm inhibition zone) and no inhibition towards Escherichia coli, whose double-layered peptidoglycan membrane confers inhibition insensibility.
